Fluorescein derivatives are the most common fluorescent reagents for biological research because of their high absorptivity, excellent fluorescence quantum yield, and good water solubility.
Fluorescein-based dyes and their conjugates have several performance characteristics that may facilitate or limit use in certain applications. Fluorescein displays:

FITC一般指异硫氰酸荧光素,异硫氰酸荧光橙红,异硫氰酸荧光黄,异硫氰酸荧光红,异硫氰酸荧光素异构体I,5-异硫氰酸荧光素。其形态为黄色粉末,有吸湿性,是一种生化试剂,也是医学诊断药物,可对由地细菌病毒和寄生虫等所致疾病进行快速诊断。并且它能与各种抗体蛋白结合,结合后的抗体不丧失与一定抗原结合的特异性,并在碱性溶液中仍有强烈绿色荧光,加酸后析出沉淀,荧光消失,微溶于丙酮、乙醚和石油醚。该品是生化试剂,也是医学诊断药物,主要用于荧光抗体技术中的荧光染料,能和各种抗体蛋白结合,结合后的抗体不丧失与一定抗原结合的特异性,并在碱性溶液中具有强烈的绿色荧光。用于医学,农学和畜牧等方面,可对由地细菌病毒和寄生虫等所致疾病进行快速诊断。
(fluorescein isothiocyanate, FITC) FITC纯品为黄色或橙黄色结晶粉末,易溶于水和酒精溶剂。有两种异构体,其中异构体Ⅰ型在效率、稳定性与蛋白质结合力等方面都更优良。FITC分子量为389.4,更大吸取光波长为490~495nm,更大发射光波长为520~530nm,呈现明亮的黄绿色荧光。FITC在冷暗干燥处可保存多年,是目前应用最广泛的荧光素。其主要优点是人眼对黄绿色较为敏感,通常切片标本中的绿色荧光少于红色。危险代码:Xn,危险等级:R20/21/22-R36/37/38-R42/43。安全等级:S22-S26- S36/37/39-S45。
